A homogenous enzyme immunoassay (EIA) for measurement of serum thyroxine (T4) concentration was evaluated for use with canine and feline serum. The EIA method was linear from 0 to 150 nmol T4/L for human serum, 0 to 94 nmol T4/L for feline serum and 10 to 60 nmol T4/L for canine serum. The objective of this experiment was to quantify ruminal digestive processes that could help to identify factors limiting DMI when silages differing in grass maturity were fed to dairy cows. Four silages were harvested at 1-wk intervals from a primary growth of a timothy-meadow fescue sward, resulting in feeds with digestible OM content in DM (D-value) of 739, 730, 707, and 639 g/kg in the order of succeeding harvest date. Four ruminally cannulated dairy cows were given ad libitum access to these silages supplemented with 7 kg concentrate per day in a 4 x 4 Latin square design. Rumen function was clearly affected by decreasing digestibility of silage fed. Passage rate of digestible NDF (DNDF) and indigestible NDF (INDF) increased, but it could not prevent the accumulation of DM, NDF, DNDF, and INDF into the rumen when silages of progressing grass maturity were fed. The greatest proportional increases in rumen pool were found in INDF and in medium particles (separated by wet sieving and measuring 315 to 2,500 microm). The passage of medium INDF particles decreased (P < 0.01) linearly (from 0.0365/h to 0.0281/h) with increasing maturity of grass ensiled, and it was slower than passage of small (80 to 315 microm) particles (on average 0.0524/h). Particle size reduction of large INDF particles to medium INDF particles was slower (P < 0.001) in the early cut silages (0.0216/h to 0.0484/h) but reduction of medium INDF particles to small INDF particles was faster (P < 0.001) in early cut silages (0.0436 to 0.0305). Passage of medium size particles and(or) rate of medium particle breakdown to small particles were potential intake-constraining properties of low digestibility forages, whereas large particle reduction to medium particles seemed not to be limiting. The increased feed intake of the early-cut silages was accompanied by decreased rumen fill, suggesting that rumen fill was not at least solely responsible for feed intake control.  相似文献

The cadmium (Cd) contents of oats (Avena sativa L.) in Finland were examined over a 3 year period in three types of trials: official variety, organic vs conventional cultivation, and nitrogen fertilization trials. Large seasonal and regional variations were found in the Cd concentrations. In official variety trials, the mean Cd contents in 1997, 1998, and 1999 were 0.046, 0.029, and 0.052 mg kg(-1) dry weight (dw), respectively, ranging from 0.008 to 0.120 mg kg(-1) dw. The concentrations were generally well below the maximum permitted level of 0.100 mg kg(-1) fresh weight. No significant differences were found between the organic and the conventional cultivation techniques. Nitrogen (N) fertilization increased the Cd contents of oats especially at high nitrogen rates (160 kg N hectare(-1) (ha)). Significant cultivar differences (p < 0.001) were determined in all trials. Cultivars Salo and Kolbu had consistently higher contents, and Belinda and Roope had lower Cd contents among the different growing conditions. Hence, it is possible to cultivate and develop oat cultivars less likely to accumulate Cd.  相似文献

While X-ray scanning is increasingly used to measure the interior quality of logs, terrestrial laser scanning (TLS) could be used to collect information on external tree characteristics. As branches are one key indicator of wood quality, we compared TLS and X-ray scanning data in deriving whorl locations and each whorl’s maximum branch and knot diameters for 162 Scots pine (Pinus sylvestris L.) log sections. The mean number of identified whorls per tree was 37.25 and 22.93 using X-ray and TLS data, respectively. The lowest TLS-derived whorl in each sample tree was an average 5.56?m higher than that of the X-ray data. Whorl-to-whorl mean distances and the means of the maximum branch and knot diameters in a whorl measured for each sample tree using TLS and X-ray data had mean differences of ?0.12?m and ?6.5?mm, respectively. One of the most utilized wood quality indicators, tree-specific maximum knot diameter measured by X-ray, had no statistically significant difference to the tree-specific maximum branch diameter measured from the TLS point cloud. It appears challenging to directly derive comparative branch structure information using TLS and X-ray. However, some features that are extractable from TLS point clouds are potential wood quality indicators.  相似文献

ObjectiveTo study the effects of oromucosal detomidine gel administered sublingually to calves prior to disbudding, and to compare its efficacy with intravenously (IV) administered detomidine.Study designRandomised, prospective clinical study.AnimalsTwenty dairy calves aged 12.4 ± 4.4days (mean ± SD), weight 50.5 ± 9.0 kg.MethodsDetomidine at 80 μg kg?1 was administered to ten calves sublingually (GEL) and at 30 μg kg?1 to ten control calves IV (V. jugularis). Meloxicam (0.5 mg kg?1) and local anaesthetic (lidocaine 3 mg kg?1) were administered before heat cauterization of horn buds. Heart rate (HR), body temperature and clinical sedation were monitored over 240 minutes. Blood was collected from the V. cephalica during the same period for drug concentration analysis. Pharmacokinetic variables were calculated from the plasma detomidine concentration-time data using non-compartmental methods. Statistical analyses compared routes of administration by Student’s t-test and linear mixed models as relevant.ResultsThe maximum plasma detomidine concentration after GEL was 2.1 ± 1.2 ng mL?1 (mean ±SD) and the time of maximum concentration was 66.0 ± 36.9 minutes. The bioavailability of detomidine was approximately 34% with GEL. Similar sedation scores were reached in both groups after administration of detomidine, but maximal sedation was reached earlier in the IV group (10 minutes) than in the GEL group (40 minutes). HR was lower after IV than GEL from 5 to 10 minutes after administration. All animals were adequately sedated, and we were able to administer local anaesthetic without resistance to all of the calves before disbudding.Conclusions and clinical relevanceOromucosally administered detomidine is an effective sedative agent for calves prior to disbudding.  相似文献

ObjectiveTo investigate plasma drug concentrations and the effect of MK-467 (L-659′066) on sedation, heart rate and gut motility in horses sedated with intravenous (IV) detomidine.Study designExperimental randomized blinded crossover study.AnimalsSix healthy horses.MethodsDetomidine (10 μg kg?1 IV) was administered alone (DET) and in combination with MK-467 (250 μg kg?1 IV; DET + MK). The level of sedation and intestinal sounds were scored. Heart rate (HR) and central venous pressure (CVP) were measured. Blood was collected to determine plasma drug concentrations. Repeated measures anova was used for HR, CVP and intestinal sounds, and the Student's t-test for pairwise comparisons between treatments for the area under the time-sedation curve (AUCsed) and pharmacokinetic parameters. Significance was set at p < 0.05.ResultsA significant reduction in HR was detected after DET, and HR was significantly higher after DET + MK than DET alone. No heart blocks were detected in any DET + MK treated horses. DET + MK attenuated the early increase in CVP detected after DET, but later the CVP decreased with both treatments. Detomidine-induced intestinal hypomotility was prevented by MK-467. AUCsed was significantly higher with DET than DET + MK, but maximal sedations scores did not differ significantly between treatments. MK-467 lowered the AUC of the plasma concentration of detomidine, and increased its volume of distribution and clearance.Conclusions and clinical relevanceMK-467 prevented detomidine induced bradycardia and intestinal hypomotility. MK-467 did not affect the clinical quality of detomidine-induced sedation, but the duration of the effect was reduced, which may have been caused by the effects of MK-467 on the plasma concentration of detomidine. MK-467 may be useful clinically in the prevention of certain peripheral side effects of detomidine in horses.  相似文献

Updating of working memory has been associated with striato-frontal brain regions and phasic dopaminergic neurotransmission. We assessed raclopride binding to striatal dopamine (DA) D2 receptors during a letter-updating task and a control condition before and after 5 weeks of updating training. Results showed that updating affected DA activity before training and that training further increased striatal DA release during updating. These findings highlight the pivotal role of transient neural processes associated with D2 receptor activity in working memory.  相似文献

The endangered Sonoran topminnow Poeciliopsis occidentalis has steadily declined in distribution and abundance in the past several decades, and currently survives in the United States only in several isolated localities in southern Arizona. This reduction is correlated with, and primarily attributed to, habitat destruction, and introduction and establishment of mosquitofish Gambusia affinis and other exotic fishes. Topminnows have characteristically been reduced in number or replaced within a year or two of introduction of non-native fishes. Other native fishes have experienced similar declines after introduction of exotics, and much of the endemic western ichthyofauna may be vulnerable to extirpation in this manner. Predation by introduced fishes on natives appears to be a likely mechanism of replacement.  相似文献
